Post-doc in lipid deuteration for neutron science

Wood decomposition represents a major process in the cycles of carbon and nutrients in terrestrial ecosystems. Brown-rot fungi are decomposers with the unique ability to break down wood carbohydrates without substantial lignin removal, which has enormous implications for the carbon quality that enters soils in coniferous forests where brown-rot fungi are prevalent. Brown-rot involves a combination of non-enzymatic and enzymatic decomposition of wood with iron playing a central role in the non-enzymatic process. Iron chemistry, its mobilization and translocation by the fungi during wood decay, however, remain poorly understood. Furthermore, the effect of the non-enzymatic reactions on carbohydrate and lignin decomposition require more attention. This project will examine changes in iron oxidation state and translocation, and changes in the chemical and structural characteristics of wood components during colonization of wood sections by brown-rot fungi under laboratory conditions. This will be done by employing a series of cutting-edge techniques including synchrotron-based X-ray microscopy, atomic force microscopy-infrared spectroscopy, and nanoscale secondary ion mass spectrometry, providing an unprecedented spatial resolution for tracing the reactions directly where they occur and clarifying the mechanisms driving brown-rot decay. The results of the project will contribute to a deeper understanding of fungal ecology with implications for applications connected to wood utilization and preservation.
